The main way to get characters is to play survival.
If you get far enough with certain characters, you'll fight someone you don't have yet.
Example: I got to fight 21 with Sasuke and fought Itachi. After I lost, I could use Itachi.
Other Example: Use Garaa to unlock Baki, Use Naruto to unlock Kabuto.

Go to Time Attack, and get a time of less then 8 minutes. After you are done with the tenth battle, the credits will play and a caption will say that you have unlocked Difficulty Level 4.

Get first place in Score Attack mode to unlock Kumite mode.
Survival mode
Get first place in Time Attack mode to unlock Survival mode.
Bonus characters
Successfully complete one of the following tasks to unlock the corresponding character. There are multiple ways to unlock each character. Note: To determine your mission completion percentage, select "Extra" at the main menu, choose "Rankings", then select "Mission Completion".
Awaken Hinata
1. Unlock all characters except for 9 Tail, Sasuke Second State, Bando, and Kagura.
2. Reach 100% combined mission completion for Hinata and Neji.
3. Reach 300% combined mission completion for all characters.
4. Fight in 700 battles.
Anko
1. Reach 15% mission completion for Orochimaru.
2. Fight in 350 battles.
Asuma
1. Successfully complete Story modes on CPU difficulty Level 2 or higher.
2. Reach 30% combined mission completion for Ino, Shikamaru, and Choji.
3. Fight in 160 battles.
Baki
1. Successfully complete Story modes on CPU difficulty Level 2 or higher.
2. Reach 30% combined mission completion for Gara, Temari, and Kankuro.
3. Fight in 350 battles.
Bando
1. Successfully complete Story modes on CPU difficulty Level 2 or higher.
2. Unlock all characters except for Nine Tailed, Sasuke Second State, Kagura, and Hintata.
3. Reach 20% mission completion for Kabuto.
Itachi

Kagura

Kisame
1. Reach 30% combined mission completion for Guy, Asuma, and Kurenai.
2. Fight in 300 battles.
Komachi
1. Successfully complete Story modes on CPU difficulty Level 2 or higher.

Kurenai
1. Successfully complete Story modes on CPU difficulty Level 2 or higher.
2. Reach 30% combined mission completion for Hinata, Kiba, and Shino.
3. Fight in 120 battles.
Orochimaru
1. Reach 20% combined mission completion for Jiraiya and Tsunande.
2. Fight in 200 battles.
Sasuke Second State
1. Unlock all characters except for Nine Tailed, Bando, and Kagura.
2. Reach 50% mission completion for Sauaske.
3. Reach 300% combined mission completion for all characters.
4. Fight in 800 battles.
Towa
1. Successfully complete Story modes on CPU difficulty Level 2 or higher.
2. Fight in 80 battles.
Nine Tailed
1. Unlock all characters except for Sasuke Second State, Bando, and Kagura.
2. Reach 50% mission completion for Naruto.
3. Reach 300% combined mission completion for all characters.
4. Fight in 750 battles.
Yugao Uzaki
1. Successfully complete Story modes on CPU difficulty Level 2 or higher.
2. Reach 30% combined mission completion for Baki, Komachi, and Towa.
3. Fight in 450 battles.

Baki: Beat Gaara, Kankuro, and Temari's first 2 missions with jonin rank or higher.

Ultimate Nine-Tailed Naruto: Beat Naruto's first 3 missions wiht hokage rank, beat Naruto's 1-on-3 Battle! mission and his Two man squad! mission with head ninja rank or higher, if he is still nt unlocked lose a few more of Naruto's missions and he should appear.
Sasuke Second State: Same as Ultimate Nine-Tailed Naruto except do it all with Sasuke.
Awakened Hinata: Same as Ultimate Nine-Tailed Naruto except do it all with Hinata then do it all with Neji.
